چکیده

Binding of the triazinone derivative metribuzin to thylakoid membranes was studied in depend ence of tem perature and trypsin pre-treatm ent of the chloroplasts. The tem perature curve of the binding constant suggests that the lipid phase of the membrane contributes to inhibitor binding. Trypsin pre-treatm ent of the membranes removes the receptor sites for the inhibitor, while the trypsin effect on the affinity of the receptor to the inhibitor is marginal. Concomitantly trypsin treatm ent releases the inhibitory effect of metribuzin on the electron transport from DPC to DCPIP.

Characterization of the Inhibition of Photosynthetic Electron Transport in Pea Chloroplasts by the Herbicide 4,6-Dinitro- o-cresol by Comparative Studies with 3-(3,4-Dichlorophenyl)-l,l- dimethylurea

1. The percent inhibition of oxygen evolution by a given herbicide concentration is the same at various light intensities except at very low intensities where the percent inhibition becomes larger. The same results are obtained with the herbicide DCMU. 2. The concentration of DCMU causing 50% inhibition of oxygen evolution decreases with de creasing chloroplast (and thus of chlorophyll) concen...

Stimulation of Photosystem I Electron Transport by High Concentration of 3-(3,4-Dichlorophenyl)-1,1-dimethyl Urea in Uncoupled Chloroplasts.

The light saturated rate of photosystem I-dependent electron transport (ascorbate/dichlorophenol-indophenol --> methyl vilogen in presence of 1 micromolar 3-[3,4-dichlorophenyl]-1,1-dimethyl urea [DCMU]) was increased by a high concentration of DCMU added to broken and uncoupled chloroplasts isolated from pea (Pisum sativum). At 50 micromolar DCMU, the increase was around 50%. No stimulation wa...

Inhibition of Photosynthetic Electron Flow by Phenol and Diphenylether Herbicides in Control and Trypsin-Treated Chloroplasts

Alkyl substituted 2-halogen-4-nitro-phenols (like bromonitrothymol) are potent inhibitors of photosynthetic electron flow in chloroplasts. There inhibition site is identical to that of DCMU, though the phenols do not contain their essential chemical element responsible for inhibition nor do they follow their correlation of chemical structure and biochemical function. Binding of labelled metribu...

Comparison of the Inhibition of Photosynthetic Reactions in Chloroplasts by Dibromothymoquinone, Bromonitrothymol and Ioxynil

The inhibition of photosynthetic electron flow in broken chloroplasts by dibromothymoquinone and dibromothymohydroquinone (DBMIBH2) is reversed by dithiothreitol (DTT) as well as by serum albumin. The reversal of DBMIBH2 inhibition by DTT shows a time lag, that of DBMIB only, when chloroplasts and DBMIB had been preilluminated. This is to show that chloroplasts reduce DBMIB and that probably DB...

Dichlorophenylurea-insensitive Reduction of Silicomolybdic Acid by Chloroplast Photosystem II.

The photoreduction of silicomolybdate and other heteropoly ions by chloroplasts is insensitive to 3-(3, 4-dichlorophenyl)-1, 1-dimethylurea (DCMU). Both water and diphenylcarbazide can be used as electron source for the reduction. Three different assays for silicomolybdate reduction are described including oxygen evolution, formation of a reduced heteropoly blue silicomolybdate, or an indirect ...
